Following technique uses radioactive precursors for observing metabolic activities of macromolecules, is:

    A)  chromatography

    B)  density gradient centrifugation or cell  fractionation

    C)  autoradiography

    D)  electron microscope

    Correct Answer: C

    Solution :

Autoradiography uses radioactive precursors for observing metabolic activities of macromolecules. A specimen is exposed to a solution containing radioactive molecules like Tritium\[({{H}^{3}})\], Carbon \[({{C}^{14}})\], Phosphorus\[({{p}^{32}})\] and than autoradiography is used to locate radioactive isotopes in cells, tissues and organs. Chromatography (chroma-colour-graphy- writing) is a new technique which was invented by Michael Tswett. The technique is employed for the separation of constituents of mixture and based upon the principle of partition coefficient. Density gradient centrifugation is used to separate cell organelles from cell homogenate through a gradient of increasing sucrose density. Electron microscopy is the best method available to study ultrastructure of a cell. The resolving power is 0.3 - 0.5 nm and final magnification can be 106 times or more.
