Which of the following is used for observing spindle fibres?

    A)  Dark Held microscope

    B)  Phase contrast microscope

    C)  Polarisation microscope

    D)  Scanning transmission electron microscope

    Correct Answer: B

    Solution :

Phase contrast microscope is helpful to study the physical changes produced during various cellular events such as spindle. formation, karyokinesis, cytokinesis, cyclosis, pinocytosis, phagocytosis, oogenesis, spermatogenesis, secretion of glandular cells etc. In this microscope an enhancement is seen in the contrast between cells and their environment along with internal organelles and their surroundings due to difference in densities of these objects. Dark field microscope is used to see those objects which do not refract light. This microscope is particularly valuable for organisms which stain poorly. Polarisation microscope is based upon the behaviour of certain cell and tissue components, when they are observed with polarized light. Scanning electron microscope gives a surface view of structures by forming an image with the secondary electrons reflected by those structures. It is specially useful in studing the surface structures of intact cells and viruses.
