                     DNA ligase (Khorana 1967) is the enzyme used during DNA replication for joining Okazaki segments. The strand formed by joining Okazaki fragments is called lagging or discontinuous strand. DNA ligase is also used in recombinant DNA technology. Restriction endonucleases are used to break DNA molecule into smaller segments. These enzymes are obtained from bacteria. They break DNA at specific sites called restriction sites. DNA polymerase was discovered by Komberg in E. coll. This enzyme is responsible for the polymerization of deoxyribonucleoside triphosphate on a DNA template to form a new complementary DNA strand. They are of 3 types, DNA polymerase-I, DNA polymerase-II and DNA polymerase-III. Gyrase is present in prokaryotes, it is responsible for untwisting of DNA.
